02-P011 Identification of new cell growth regulators in Drosophila melanogaster by genome wide RNAi screening

چکیده
the A8, and it has been shown that the gene double sex (dsx) represses growth in the former, probably by repressing decapentaplegic (dpp) expression. We show here that the absence of Abd-B or homothorax (hth) in this segment also increases growth and activates the expression of dMyc and dpp. The effects on the size of the A9 can be rescued if dpp activity is reduced. Neither Abd-B, dsx, nor hth regulates each other’s expression, which suggest that dsx, hth and Abd-B may coordinately repress growth in this primordium of the genital disc.

Genome-scale RNAi on living-cell microarrays identifies novel regulators of Drosophila melanogaster TORC1-S6K pathway signaling.
The evolutionarily conserved target of rapamycin complex 1 (TORC1) controls cell growth in response to nutrient availability and growth factors. TORC1 signaling is hyperactive in cancer, and regulators of TORC1 signaling may represent therapeutic targets for human diseases. To identify novel regulators of TORC1 signaling, we performed a genome-scale RNA interference screen on microarrays of Dro...
متن کاملAnalysis of neural stem cell self-renewal and differentiation by transgenic RNAi in Drosophila.
The fruit fly, Drosophila melanogaster, has proved to be a useful model organism for studying the biology of neural stem cells. Notably, significant progress has been made in identifying the molecular mechanisms that regulate the asymmetric cell divisions of the neural stem cell-like neuroblasts during brain development. Recently, the emerging technology of genome-wide transgenic RNA interferen...
متن کاملModulation of human JAK-STAT pathway signaling by functionally conserved regulators
Both the core JAK-STAT pathway components and their in vivo roles have been widely conserved between vertebrates and invertebrate models such as Drosophila melanogaster. Misregulation of JAK-STAT pathway activity has also been identified as a key factor in the development of multiple human malignancies. Recently, whole genome RNA interference (RNAi) screens in cultured Drosophila cells have ide...

Background: Flubendiamide, comparatively a new pesticide designed to eradicate lepidopteran insect pests is known to have low risk to birds, mammals, fish, algae, honey bees, non-target arthropods, earthworms, soil macro- and micro-organisms, non-target plants as well as sewage treatment organisms; however, the risk assessment for aquatic invertebrates from metabolite could not be finalized wit...
متن کاملRealizing the promise of RNAi high throughput screening.
Recently reporting in Nature, Collinet et al. describes the application of quantitative multiparametric methods to a genome-wide RNAi screen for regulators of endocytosis. The study illustrates the power of this approach beyond the identification of new endocytic components to providing insights into the design principles of the endocytic system.
